Immobilization of bacteria and Saccharomyces cerevisiae in poly(tetrafluoroethylene) membranes.
A novel method for immobilization of bacteria and Saccharomyces cerevisiae cells is described. Microorganisms may be entrapped in a matrix of poly(tetrafluoroethylene) (PTFE) fibrils. Cells are blended with an aqueous emulsion of PTFE stabilized with Triton X-100 surfactant to form a thick paste. The paste is calendered biaxially in a standard rubber mill. This process causes fibrillation of th...
متن کاملBiosynthesis of natural flavanones in Saccharomyces cerevisiae.
A four-step flavanone biosynthetic pathway was constructed and introduced into Saccharomyces cerevisiae. The recombinant yeast strain was fed with phenylpropanoid acids and produced the flavanones naringenin and pinocembrin 62 and 22 times more efficiently compared to previously reported recombinant prokaryotic strains. Microbial biosynthesis of the flavanone eriodictyol was also achieved.

Epitope Tagging of Strains Regulators were tagged by inserting multiple copies of the Myc epitope coding sequence into the normal chomosomal loci of these genes. The plasmid pWZV88 (1) was used as a template to generate PCR products containing the Myc epitope coding sequence and a selectable marker (TRP) flanked by homologous regions designed to recombine at the 3’ end of the targeted transcrip...
متن کاملComparative Genomics of Saccharomyces cerevisiae Natural Isolates for Bioenergy Production
Lignocellulosic plant material is a viable source of biomass to produce alternative energy including ethanol and other biofuels. However, several factors—including toxic by products from biomass pretreatment and poor fermentation of xylose and other pentose sugars—currently limit the efficiency of microbial biofuel production. To begin to understand the genetic basis of desirable traits, we cha...
متن کاملIncipient Balancing Selection through Adaptive Loss of Aquaporins in Natural Saccharomyces cerevisiae Populations
A major goal in evolutionary biology is to understand how adaptive evolution has influenced natural variation, but identifying loci subject to positive selection has been a challenge. Here we present the adaptive loss of a pair of paralogous genes in specific Saccharomyces cerevisiae subpopulations. We mapped natural variation in freeze-thaw tolerance to two water transporters, AQY1 and AQY2, p...
